Short version: yes, it does, but not nearly as dramatically as forum threads make it sound. A 250k volume pot makes a guitar a touch warmer and rounder. A 500k pot leaves it brighter and more open. On the bench I can measure the difference, and a careful player can hear it, but we are talking about a couple of decibels at the top end, not a night-and-day transplant. If your guitar sounds wrong, the pot value is usually a fine-tuning detail, not the main culprit.
Here is what the number actually means, why single-coil guitars ship with 250k and humbucker guitars with 500k, and when it is genuinely worth swapping.
What a volume pot actually does to your sound
A potentiometer is just a variable resistor. The number on it, 250k or 500k, is its maximum resistance in ohms: 250,000 or 500,000. When your guitar is dimed at 10, that pot is not out of the circuit. It is still sitting there as a fixed resistance to ground, quietly bleeding off a slice of your signal.
Every magnetic pickup has a natural resonant peak, a bump in its frequency response that gives the guitar its voice and sits up in the treble region. The resistance hanging across the pickup acts as a load that tames that peak. A lower resistance is a heavier load, so it pulls the peak down and rolls off a little high end. A higher resistance is a lighter load, so the peak stays taller and the guitar reads brighter. That is the whole mechanism in one sentence.
So a 500k pot does not "add" treble. It simply gets out of the way and lets more of the pickup's own high end through. A 250k pot removes a bit of that shimmer. Everything downstream, your cable, your amp's input, adds a little more loading on top, which is why a long coiled cable makes a bright guitar sound darker for exactly the same reason.
Why single coils get 250k and humbuckers get 500k
This is not a rule someone invented to be annoying. It is voicing.
Single coils are bright and articulate by nature, with a resonant peak that already sits high. Load them lightly with a 500k pot and that peak can tip over into ice-pick territory, especially on the bridge pickup. Fender settled on 250k decades ago because it gently rounds off the top and keeps a Strat or Tele from getting shrill. Put a scratchy old Strat next to a fresh one and the pot value is part of why one is sweet and one is glassy.
Humbuckers are the opposite problem. Two coils wired together cancel hum, but they also cancel some of the highest frequencies, so a humbucker's peak sits lower and warmer to begin with. Load that with a 250k pot and you smother it, muddy, dark, a little lifeless. Gibson uses 500k precisely to keep humbuckers open and clear. P-90s, which are single coils but fat and dark, usually get 500k for the same reason.
Here is a diagnosis I run into constantly on the bench: a player says a guitar with humbuckers sounds muddy and blames the pickups. Half the time someone has dropped 250k pots into it, or an import came from the factory that way to save a nickel. Swap in 500k and the "bad" pickups suddenly wake up. Active pickups like EMGs are their own world and use 25k pots, because the onboard preamp sets the tone and the pot is only there to control level.
So how big is the difference, really?
This is where I part ways with the internet. Changing a pot value shifts the resonant peak by roughly 2 to 3 dB. That is audible in a quiet room, on a clean amp, if you are listening for it. It is not audible across a loud stage, through distortion, in a mix, or a week later from memory. If you have ever done the swap and thought "huh, maybe a little brighter," that is the honest result, not a failure of your ears.
What that means practically: pot value is a seasoning, not a main ingredient. Pickups, string gauge, your amp and how you set it, and your hands all move the tone far more than 250k versus 500k. Reach for a pot swap when a guitar is close but consistently a hair too dark or too bright, not when you are chasing a completely different sound.
The wildcard nobody mentions: pot tolerance
Here is the detail that resolves half the arguments online. A pot labeled 500k is not exactly 500k. Standard tolerance is plus or minus 10 percent for a decent CTS pot, and up to 20 percent for cheap ones. So a "500k" pot can measure anywhere from about 400k to 600k, and a "250k" from 200k to 300k.
Do the math and you will see the ranges nearly touch. A hot 250k pot at 300k and a cold 500k pot at 400k are not that far apart. This is a real reason two guitars off the same rack can sound subtly different, and why some players swear a pot swap "did nothing" while others hear a clear change. They were not measuring the same parts. If you care, put a multimeter on the pot before you decide the value is your problem.
Should you actually swap your pots?
My honest take: leave them alone unless you have a specific complaint. If a humbucker guitar sounds muddy and you confirm it has 250k pots, yes, going to 500k is one of the best-value upgrades there is, a few dollars in parts and a bit of soldering. If a single-coil guitar is harsh and you want to soften it, 250k is the move, though a tone-capacitor change or just rolling the tone knob back is cheaper and reversible.
But if you are happy with how a guitar sounds, do not go chasing 2 dB. That way lies a parts drawer full of pots and a guitar that is always half-apart on the bench. Get the pickups and setup right first. Pot value is the last five percent, not the first fifty.
